To explore the influences of TFAM knockdown on the cardiotoxicity of doxorubicin(DOX) in experimental mice.
Doxorubicin was intraperitoneally injected into TFAM knockdown and wild typemice, and the survival rate was recorded. The variation of creatine kinase (CK) in serumwas detected using a CK activity detection kit. The apoptosis of cardiomyocytes was measured by TUNEL reagent. The change of mitochondria DNA (mtDNA) copy number was measured by quantitative PCR. The survival of mice treated with Huangqi was recorded.
The injection of DOX led to a significant higher mortality in TFAM knockdown mice, than that in wild type controls. After DOX treatment, the serum CK level and cardiomyocytes apoptosis rate were higher in TFAM knockdown mice. However, the mtDNA number was lowest in TFAM knockdown mice treated with DOX. Administration of Huangqi could not protect TFAM knockdown mice from the cardiotoxicity of DOX.
In vivo, knockdown of TFAM enhances the cardiotoxicity of DOX.
