Discussion on effect of NLRP3 inflammasome signal pathways by different exercise preconditionings on exhaustion exercise rats.
Sixty SD rats were randomly divided into four groups: control group(C), exhausive exercise group(EE), 3 days exercise preconditioning plus exhausive exercise group(3dEP＋EE), 3 weeks exercise preconditioning plus exhausive exercise group(3wEP＋EE). A light microscope and electron microscope were used to observe the pathological changes in heart tissues, the levels of IL-1β,IL-18, IL-6, hs-CRP, CK-MB, BNP in serum were detected by enzyme linked immunosorbent assay(ELISA).The levels of protein of NLRP3 and Caspase-1 were detected by Western Blot. Using the correlation analysis the relationship aong of serum IL-1 β, IL-18, IL-6, hs-CRP, BNP, CK-MB and their relation with the NLRP3, Caspase-1 protein expressions.
Under the light microscope and electron microscope, EE group heart tissues were stained obviously uneven, a large number of myocardial fibers were fractured and messed, interstitial fibers were hyperplasiaed and edemaed moderately, much of myocardial cells were edemaed and necrosised, inflammatory cells were infiltrated, the number of average infiltration inflammatory cells was highest, mitochondria were edemaed seriously, part of the crest and a small number of membrane were blended together, fuzzy and crest were fractured, sarcomere were disordered; 3dEP＋EE and 3wEP＋EE rats heart tissues injuries were lessed, among the three groups, the best results being in the 3wEP＋EE group. Compared with the normal control group, the serum contents of IL-1β, IL-18, hs-CRP, CK-MB and protein expressions of NLRP3 and Caspase-1 in EE group rats heart tissues were significantly increased (IL-1β:12.00±1.55 vs 18.77±1.28, IL-18: 150.74±7.09 vs 236.79±15.73, hs-CRP:107.15±14.22 vs 469.37±137.73, CK-MB:7.40±0.40 vs 15.57±0.91, NLRP3(grey value):0.161±0.06 vs 0.66±0.07, Caspase-1(grey value):0.48±0.06 vs 0.96±0.08, all P<0.01). Compared with the EE group, the serum contents of IL-1β, IL-18, hs-CRP, CK-MB and protein expressions of NLRP3 and Caspase-1 in 3dEP＋EE and 3wEP＋EE group were remarkably decreased, among the three groups, the best results being in the 3dEP＋EE group (IL-1β:16.57±1.81 vs 14.26±1.42, IL-18: 191.92±14.16 vs 168.49±7.05, hs-CRP: 339.56±89.65 vs 207.06±41.68, CK-MB: 13.10±0.78 vs 9.97±1.08, NLRP3(grey value):0.48±0.06 vs 0.33±0.07, Caspase-1(grey value):0.79±0.02 vs 0.65±0.06, all P<0.01). There is a positive correlation among IL-1β, IL-18, IL-6, hs-CRP, BNP, CK-MB, NLRP3, Caspase-1: (IL-1β: r=0.718, 0.756, 0.660, 0.554, 0.628, 0.728, 0.624, P<0.01; IL-18(ng/ml):r=0.858, 0.786, 0.735, 0.865, 0.813, 0.857, P<0.01; IL-6: r=0.827, 0.801, 0.839, 0.803, 0.829, P<0.01; hs-CRP: r=0.818, 0.824, 0.860, 0.801, P<0.01; BNP: r=0.823, 0.749, 0.784, P<0.01; NLRP3: r=0.893, P<0.01).
Exercise preconditioning alleviate the myocardial injury through regulate the expression of NLRP3 inflammasome signal pathways and reduce inflammatory reaction, the best results being in the 3dEP＋EE group.
