This study was designed to determine the effects of Shenfu yixin formula (SFYX) on myocardial apoptosis and fibrosis of left ventricular at the early stage of myocardial infarction (MI) in rats.
MI was induced by the ligation of left anterior descending coronary artery in adult male Sprague-Dawley rats. 24h later after operation, The rats were divided into five groups: sham-operated group; MI+vehicle group; SFYX -1.76g (1.76g/kg/day) group; SFYX -8.80g (8.80g/kg/day) group and losartan (10mg/kg/day) group.The rats in each grope were administrated for 1 week by oral gavage once a day after MI. The left ventricular structure and function were measured by echocardiography, and the level of myocardial apoptosis and collgen were determined by TUNEL method and masson＇s trichrome staining .
Compared to sham-operated group,left ventricular end-diastolic dimension (LVEDD,0.88±0.03cm) and left ventricular end-systolic dimension (LVESD,0.70±0.02cm) were increased and left ventricular ejection fraction (LVEF,43.25±3.60%) and left ventricular fractional shortening (LVFS,20.68±2.81%) were decreased in MI+vehicle group(P<0.05);SFYX (8.80g/kg/day) and losartan reduced LVEDD(0.51±0.02cm) and LVESD(0.29±0.02cm) and increased LVEF(79.36±2.56%) and LVFS(41.50±2.05%) compared with MI+vehicle(P<0.05). Pathological results showed that the level of myocardial apoptosis and collgen were obvious in each group except sham-operated group.But myocardial apoptosis index(MAI) and collgen volume fraction(CVF) of Ischemia dangerous area were reduced in SFYX -8.80g group (1.8±0.07%,13.5±2.8%) and Losartan group (1.2±0.04%, 12.1±2.7%) compared to MI+vehicle group(7.5±0.4%, 22.2±3.2%)(P<0.05).
Our date showed that treatment with SFYX after MI improved cardiac function,and reduced the level of myocardial apoptosis and collgen.The results suggest that SFYX attenuated MI induced left ventricular remodeling possibly via inhibition of myocardial apoptosis.
