Background: Insulin resistance(IR) is an established risk factor for hypertension (HT). Calpain-10 (CAPN10) participates in glucose uptake pathways in skeletal muscle and reduced CAPN10 expression in muscle cells leads to a reduction in insulin-mediated glucose uptake. Reduced skeletal muscle CAPN10 mRNA content was reported in carriers of risk genotype (G/G) at single nucleotide polymorphism(SNP)-43 within intron 3 of the CAPN10 gene, which is thought to be candidate for IR. However, it was recently reported that GA/AA genotype of SNP- 43 was related to IR in colorectal cancer, and that AA genotype showed elevated IR in polycystic ovarian syndrome. The aim of this study was to examine the association between HT and SNP-43 of CAPN10 gene in a large-scale cross-sectional study of Japanese.
Methods: We analyzed 19278 Japanese subjects, including 8718 HT cases(SBP≥140 or DBP≥90mmHg) and 10560 non-HT controls, all of whom enrolled in the genome banking project of the 21st Century COE (Center of Excellence) Program at Jichi Medical University. All subjects were divided into four groups according to gender and age (≤49, ≥50). Genotypes (GG/GA/AA) of SNP-43(rs3792267) on CAPN10 were determined by the TaqMan PCR method in Jichi Medical University. Because the incidence of AA was low(<1.5%), GA and AA were combined together as A carrier in the following analyses. We compared the frequencies of genotypes between cases and controls by chi-square test. The strength of the associations was estimated by odds ratios(ORs) and 95% confidence intervals(CI) by using logistic regression analysis. The ORs were adjusted for age and BMI.
Results: The distributions of GG and A carrier significantly differed between cases and controls in F≤49 (p=0.040). Compared to GG, the ORs (95%CI) of A carrier were 1.16(0.85-1.57) in M≤49, 1.42(1.02-1.98 p=0.04) in F≤49, 1.04(0.88-1.24) in M≥50, and 1.06(0.91-1.24) in F≥50.
Conclusions: Compared to GG, A carrier on CAPN10 (rs3792267) may significantly increase the risk of HT in Japanese females aged≤ 49 years.
