To explore the effect of Shen Yuan Dan(SYD) on hippocampal formation, in the Sprague-Dawley(SD) rats of acute myocardial infarction(AMI)/depression, and study the mechanisms of the theory “invigorating qi &dispersing stasis” in AMI/depression.
30 SD rats were allocated into normal group, model group,H-SYD group,L-SYD group and control group randomly. The model was established by coronary artery ligation with chronic mild unpredictable stimulation and feeding alone. After 18 day's pretreatment with large dose of SYD, low dose of SYD, fluoxetine or distilled water. All the rats were sacrificed 3 days later. Light microscope(40*10)was used to observed the tissue structure of hippocampus(CA1&CA3).
Compared with rats in normal group, rats in model group showed that structure of hippocampal was obviously destroyed. Cell layer was markedly reduced and diluted. Pyramidal neurons and granular cell bands arrayed in disorder, loose. Neuronal membrane ruptured. The nucleus became smaller. Cells became atrophy. The neuronal vacuolization and pyknosis were evident, and a large number of cells were lost, and stained to observe the morphology of the neuronal nissl bodies even dissolved. The structure of hippocampal in SYD group and control group were basically complete. The arrangement of cells was regular and the layers were rich. The neuronal membrane was intact in main. Only a small part of nucleus became atrophy, vacuolization and pyknosis.
In this study,SYD can promote the repair of cell injury in hippocampal as fluoxetine. Indicating that Shen Yuan Dan can antagonize stress injury and induce newborns of hippocampal neurons, reversing the occurrence of hippocampal neuronal apoptosis.
