To evaluate the condition of gene transfection in non-targeted tissue When the Ultrasound targeted microbubble destruction(UTMD) technique mediated exogenous Bone morphogenetic protein 2 (BMP2) gene transfer to the heart in rats with acute myocardial infarction(AMI).
44 SD rats with AMI were randomly divided into 4 groups:Simple ultrasound irradiation group(M+U,n=10); Ultrasound irradiation + carrying pAd-ERFP/BMP2(The biotin recombinant adenovirus expressing enhanced red fluorescent protein and BMP2.) gene-microbubble administration 1 day group(M+S1+U,n=10), continuous administration two days group(M+S2+U,n=10). On the eighth day of the operation, The expression of ERFP in liver, lung and kidney were observed by laser scanning confocal microscopy.
Expression of ERFP in liver, lung and kidney: there was a small amount of expression in the drug delivery group and no expression in the control group. The difference between the drug delivery group and the control group was statistically significant(P<0.05), but there was no significant difference between the different drug delivery groups.
When the UTMD technique mediated exogenous BMP2 gene transfer to the heart in rats with AMI, non target tissues will also be transfected. But the transfection of non target tissue was related to the administration of BMP2, which was not related to the dosage.
