Cholesterol loading induce Vascular smooth muscle cells (VSMCs) damage and this change may play an important role in the progression of atherosclerosis. However, the mechanism of it are still not known.
In the present study, the impact and the mechanism of cholesterol loading on VSMCs were observed. Cell apoptosis, monocyte adhesion and NF/kB pathway relative proteins expression were detected by using the ANNEXIN/7AAD staining, fluorescence microscope and western blot, respectively.
Our data show that cell apoptosis and monocyte adhesion on VSMC were significantly increased in cholesterol loading VSMCs. Meanwhile, cholesterol loading activated TLR4-MyD88-p65 pathway in VSMCs and NF/kB inhibitor PDTC abolished these effects of cholesterol.
Cholesterol loading promotes VSMCs apoptosis and monocytes adhesion on it due to activation NF/kB pathway.
