This study was designed to investigate the effect of Bushen Kangshuai Tablet on the formation and development of atherosclerosis in apolipoprotein E knockout (apoE-/-) mice fed with a Western type diet, and to explore its possible mechanism of action.
C57BL/6J wild type mice as control group were fed normal diet. ApoE-/- mice fed a Western type diet were randomly divided into: model group, Bushen Kangshuai group, simvastatin group. After 4, 8, 12 weeks respectively, 8 mice of each group were selected randomly to sacrificed for dynamic observation of the indexes. A series of assays were used to detect the effect and mechanism of Bushen Kangshuai Tablet on spleen weight, spleen index, level of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) and Angiotensin II (Ang II) in serum, morphology of aortic sinus and spleen, expression of Th1 related factors granulocyte-macrophage colony stimulating factor (GM-CSF), T-bet and Interferon-γ (IFN-γ).
Compared with model group, Bushen Kangshuai Tablet reduced the degree of atherosclerotic lesions at 4, 8, 12 weeks. Moreover, Bushen Kangshuai Tablet increased HDL-C level, reduced Ang II level, spleen weight and spleen index, relieved splenomegaly and inhibited the expression of GM-CSF, T-bet, IFN-γ after 12 weeks.
Bushen Kangshuai Tablet has an inhibiting effect on the formation and development of atherosclerotic plaque. Its mechanism may be related to improve lipid metabolism, reduce Ang II level in serum and regulate spleen Th1 adaptive immune pathway.
